Molecular DNA markers in red clover breeding

The paper presents the results of the North Caucasus Research Institute of Mountain and Foothill Agriculture-a branch of the Federal State Budgetary Institution of Science of the Federal Center "Vladikavkaz Scientific Center of the Russian Academy of Sciences". Studies on the formation of complex hybrid populations, the assessment of collection samples of clover marker-oriented selection on the complex of economically valuable characteristics for the creation of varieties of hay-pasture type is given. For the study, we studied promising plants of meadow clover, the seeds of which were sown in a wide row and individually in a row. According to the results of PCR analysis of meadow clover using SSR primers, it was revealed that the Wild sample has a genetic similarity with the varieties Tetraploid VIC and Veteran when using 3 pairs of microsatellite primers (RCS 0017R/F; RCS 1307R/F and RCS 3711R/F). Also, the Tetraploid VIC and Veteran varieties have proven themselves based on the results of long - term observations, as they are characterized by high winter hardiness-up to 90%, resistance to various clover diseases and high longevity, and the Wild-growing sample can be considered promising in breeding for breeding a long-lived and stable variety in pasture use. The Vladikavkazsky variety is similar to the resistant varieties only in 2 pairs of primers (identical fragments of amplified DNA with the Tetraploid VIC and Veteran varieties with the RCS 1640R/F primer, as well as with the Veteran variety with the RCS 1307R/F primer). PCR analysis using the RCS 4001R/F primer revealed a genetic similarity of the Vladikavkazsky variety with the Early 2 variety, which in multiple field experiments showed a low level of longevity.
