The effect of the PCV-2 on the main lymphocytes and macrophages populations in the infected weaning piglets’ bronchial lymph nodes tissues

PCV-2 Swine disease is widespread in domestic and foreign pig farms. This disease IS characterized by a variety of clinical signs and pathological changes, often similar to other diseases of pigs, which explains the need for the integrated approach to diagnosis. Circovirus disease of pigs is characterized by lymphoid organs pathology, for the detection of which pathomorphological and histological studies methods are used. To determine the quantitative and qualitative changes in different populations of immunocompetent cells in tissues, the immunophenotyping method is used. This study presents a method of lymphocytes immunophenotyping in tissues and organs in pigs, based on the method of indirect immunohistochemical analysis. Pathological material for the study was selected from healthy and sick weaning piglets 60-120 days. The study was performed on native histological sections of bronchial lymph nodes made by cryotomy. When performing indirect immunohistochemical analysis, mouse monoclonal antibodies to markers of CD3, CD4 and CD8 -T-lymphocytes, CD20 - B-lymphocytes and CD14 - human macrophages were used, which made it possible to establish the possibility of interspecific cross-reactivity this antibodies used with markers of immunocompetent pig cells. These studies have shown that in the case of porcine circovirus disease, a sharp decrease in CD3+ T-lymphocytes, CD4+ T-helper cells, CD8+ cytotoxic T-cells, CD20+ B-lymphocytes and CD14+ macrophages occurs in the tissues of the bronchial lymph nodes of weaned piglets, leading to the development of secondary immunodeficiency in naturally infected PCV-2 weaning piglets.
