Using polymorphic microsatellite markers to study the genetic diversity of Triticum aestivum L

The results of microsatellite analysis of the spring wheat genome are presented. The greatest level of polymorphism information content (PIC) has been determined for the loci identified using the primers: cfd76 (PIC=0,90), gdm33 (PIC=0,87), Xgwm155 (PIC=0,85), Xgwm337 (PIC=0,90), Xgwm437 (PIC=0,86); and this enables clear differentiation of wheat breeding material. It has been determined thatall nuclear microsatellite loci included in the analysisdisplay genetical variabilityin the studied wheat samplesthat allows recommending theirusefor identification of the crop genotypes. Based on the revealed alleles, the template of the investigated common wheat samples’ genetic affinity has been calculated, clusters have been constructed, genetic distance (Euclidean, Minkovsky) has been calculated, and genetical passports have been made according to PAST algorithm. Genetical distances between parent materials of spring wheathave been calculated; the greatest value being 5.29. Taking into accountremoteness of initial forms,to produce highly productive hybrids of common spring wheat, parent pairs have been suggested taking into account remotenessof initial forms’ relationship.
Keywords: Common wheat, microsatellite loci, primers, polymorphism, PCR-analysis, genetic distances, intraspecific hybrids.
