Production of androgenic rice digaploids with sub1a and snorkel genes resistant to prolonged flooding

The object of the study was 265 rice samples obtained in the laboratory of rice breeding and seed production at the Donskoy Research Center from crossing domestic varieties with Asian donors of flood resistance genes. The locus of resistance to flooding Sub1A was transferred from the following sources: Br-11, CR-1009, Inbara-3, TDK-1, Swarna, IR-64. The gene of vigorous growth of the Snorkel was transferred from the varieties Khao Hlan On, Ma-Zhan Red, Kharsu 80A. The objective is a comprehensive assessment of rice tolerance to prolonged loading into water and the ability of vigorous growth from under the water layer. Laboratory studies were carried out using physiological, genetic, and biotechnological methods. When rice was germinated in test tubes, samples with minimal stem growth were isolated under anaerobic conditions, which indicates the presence of the Sub1A gene in them. The smallest sprout length (3.23 cm) was observed in the sample: 4395 (Inbara-3 ½ Innovator), carrying the Sub1A locus in its genotype. The tallest samples were 4283 - 15.7 cm, and 4254 - 15.2 cm, to which the vigorous growth gene (Snorkel) was transmitted. When germinating in cylinders with a height of 42 cm, significant differences were noted between the samples in terms of the growth rate under water. On the 18th day from seed filling, the largest length of seedlings was in sample 4209 (44.0 cm), and the smallest in 4395 (3.8 cm). With the help of biotechnological methods, androgenic rice plants were obtained from four hybrid combinations. 25 green regenerating plants were grown in the greenhouse, of which 9 haploids, 11 digaploids and 5 tetraploids. A PCR analysis of these rice samples was carried out and the presence of genes for resistance to deep flooding Sub1A and Snorkel was found in some of them.
