Hematological parameters of rats under automated analysis and microscopic method

This report presents data on the hematological parameters of rats under automated analysis and the classical method - microscopy. The data on the characteristics of various indicators (hematological, biochemical, and others) in linear, SPF animals, depending on lines, gender and age, are given most fully in the literature, while the information regarding outbred ones (conventional, improved versions) is limited or fragmented. In the first series of experiments on 48 outbred rats of 5-6 months of age (25 females and 23 males) according to automatic analysis it was found that their hematological parameters did not exceed the limits of physiological norms and in most cases did not differ significantly depending on gender. However, higher levels of white blood cells and lymphocytes, a significant increase in the average red blood cell volume and hemoglobin content in them was recorded in females. In males, the average values of platelets, erythrocytes and the width of the distribution of their populations exceeded those of females. In the second series of experiments, when determining the leukogram on seven animals, it was established that the data obtained using the Vet Scan analyzer HM5 with three-component lymphocyte differentiation exceeded the visual analysis data. The error in determining the number of leukocytes and lymphocytes in the automatic mode and the visual method was 11.3-13.6%, which is consistent with the opinion of other researchers about the higher accuracy of the analyzers. However, not all of them provide full differentiation of leukocytes in many animal species. In this regard, automated hematology analyzers adapted for veterinary purposes can be used to solve monitoring and other practical issues. When conducting scientific experiments on laboratory animals, it is advisable to use them in combination with the visual (microscopic) method.
