Diagnosis Tsn1 gene of Pyrenophora tritici-repentis in wheat varieties and their resistance to races of the pathogen

Recently, Тan spot, caused by Pyrenophora tritici-repentis (Died.) Drechsler, has occupied larger area on the territory of Russia. Due to the wide spread and economic importance of this disease, the search for varieties resistant to the pathogen is of high priority. This study presents the results of the diagnosis of the functional allele of the Tsn1 gene of P. tritici-repentis, responsible for the development of necrosis (Ptr ToxA), in 27 varieties of winter wheat of Russian selection. Using the PCR method with primers for the Xfcp394 marker, the Tsn1 gene allele was found in 5 samples (18.5% of the studied amount): Agat Donskoy, Zimtra, Beauty, Marathon, Pervitsa. Under greenhouse conditions in the germination phase, the wheat varieties were evaluated for resistance to the widespread races of the pathogen in the territory of Russia. Samples of the resistant wheat were determined: 13 samples (or 48.1% of the studied amount) to the 1st race (forming two toxins: Ptr Tox A, Ptr Tox C), 27 samples (100,0%) to the 2nd race (forming the toxin Ptr Tox A), 2 samples (7.4%) to the 8th race (forming three toxins: Ptr Tox A, Ptr Tox C, Ptr Tox B). This study showed that the studied varieties possess a race-specific resistance. Analysis of the data obtained indicates that the varieties identified with PCR method as ToxA-resistant were susceptible to different races of the fungal isolate producing other toxins (ToxB and ToxC). The obtained results indicate the need for an integrated approach to the study of the resistance of wheat samples to P. tritici-repentis.
