Development of selective system in vitro for obtaining lavender callus lines resistant to low temperature

For essential oil plants, including lavender, one of the most important tasks of selection is the creation of cultivars capable to produce high yields under unfavorable environmental conditions. The biotechnological method of cell selection allows to obtain genotypes resistant to biotic and abiotic stress factors. The aim of the work is to study the effect of low-temperature stress on the growth of callus cultures to develop selective system for obtaining resistant forms of lavender. As material for research were tissues and organs of Lavandula angustifolia Mill. cultivar Stepnaya. For cell selection calluses on exponential, linear and stationary phases of the growth cycle were used. When modeling cold stress, the hardening of callus cultures (4 and 8 days), freezing (2 regimens), and thawing (4 days) was conducted. It is shown that when developing a selective system for creation low-temperature resistant lavender forms it is advisable to use calluses on the linear phase of the growing cycle. The increase in the weight of callus tissue after 4 and 8 days of hardening depended on the phase of the growth cycle. The possibility of callus freezing during 6 days when air temperature decreases down to -12оC was shown. Resistant to low temperature callus lines of L. angustifolia were obtained on the basis of developed in vitro selection method.
