Peculiarities of Melissa officinalis L. explant morphogenesis at the first phase of micropropagation in vitro

Lemon balm (Melissa officinalis L.) - a perennial herbaceous medicinal, essential oil and aromatic plant, widely used in medicine and in some sectors of economy. In FSBSI "Scientific Research Institute of Agriculture of Crimea" research on M. officinalis breeding in order to obtain cultivars with high content of essential oil and productivity are conducted. To increase the efficiency of breeding work with this valuable plant it is expedient to attract the biotechnological methods, one of which is a clonal micropropagation. The aim of study was to investigate the influence of some factors on the development of explants at the first stage of lemon balm clonal micropropagation in vitro. As a result of research the peculiarities of influence of nutrient medium hormonal composition, genotype and type of culture bottle on the development of stem segments with a node on the first stage of micropropagation of M. officinalis were revealed. Analyzing the influence of 11 variants of MS nutrient medium on the development of explants of cultivar Citronella in test tubes it was found that maximum number of shoots per explant and multiplication coefficient were on media supplemented with BAP (0,5-1,0 mg/l) and GA (0,5 mg/l). It was shown that at most experiment variants the morphometric parameters of explants development and multiplication coefficient were higher in cultivar Sobornaya in comparison with Citronella. It was noted that it is preferable to use flask (100 ml) as a culture bottle for lemon balm micropropagation. The maximum multiplication coefficients by introduction in vitro explants of Sobornaya (43,1) and Citronella (30,3) cultivars were obtained by culturing in flasks on MS medium containing 0.5 mg/l BAP. Results of the study are the basis for developing methods of M. officinalis micropropagation in vitro.
