Use of biotechnological methods in developing doubled haploids when breeding white cabbage

The prospect of using doubled haploids in accelerating breeding process is beyond doubt. In the course of studies, the composition of artificial nutrient media for the cultivation of anthers of white cabbage Brassica oleracea L. was optimized in order to increase the efficiency of morphogenetic processes and increase the yield of green regenerants. The maximum amount of morphogenic callus from the anthers of all the studied white cabbage lines was induced on a nutrient medium containing 120 g / l of sucrose and growth regulators: 3.0 mg / l of 6-BAP; 1.0 mg / l of α-NUK and 1.0 mg / l of AgNO3; 2.0 mg / L amber acid; in the anther culture in vitro, regenerative plants from 63 donor genotypes of white cabbage were developed and adapted to the ex vitro conditions. A comparative analysis of allelic state of the microsatellite locus of DNA in donor plants and regenerants was carried out with the SSR-marker BoIAB 20TR. Based on this analysis and soil control, a conclusion is drawn on the origin of the regenerants. The efficiency of methodical scheme for controlling the gamete origin of regenerants obtained in anther culture of cabbage is demonstrated, which can be used in practical breeding programs of this crop. The elements of clonal micropropagation technology of regenerants having gamete origin have been developed.
