Micropropagation of Albidae Dode section poplar hybrids

Poplar is one of the rapidly growing tree species. Introduction of selected, resistant to unfavorable environmental factors material into forestry practice is limited by low efficiency of vegetative propagation of some poplar species. The application of the in vitro culture method allows to quickly multiply valuable biotypes. In the process of micropropagation of woody plants morphogenetic reaction significantly depends on genotype, age, source material. These factors require optimization of culture conditions and the composition of culture media for specific forms, varieties, hybrids. Poplar micropropagation was carried out by activation of axillary meristems. Using isolated in June young shoots lets to obtain up to 76,3% aseptic viable cultures. The results showed that the highest percentage of morphogenic cultures was obtained using ½ WPM medium, containing BAP 0,2-0,5 mg/l. For elongation the shoots are transplanted to fresh medium every 30-35 days. The shoots 1,5-2 cm long were rooted in the hormone-free media and with the addition of auxin. The maximum rooting (50%) was achieved using IBA 0,01 mg/l as root inducer. The efficiency of all micropropagation stages is significantly depends on the genotype of the source tree. For all test forms were obtained regenerated plants useful for mass micropropagation.
